Plant Regeneration of Sweet Potato (Ipomeoa batatas L.) from Root Tissue Cultured in Vitro

Abstract
Successful plant regeneration was obtained from root tissue of four sweet potato cultivars, Suwon147, Chungsung, Jinmi, and White Star, which were grown in vitro. Regeneration ability was the best with Suwon147 followed by Chungsung, Jinmi, and White Star. Two-month-old root tissue cultured on 1/2MS medium containing 0.5 §­ NAA in dark condition was fumed out to be the best material for shoot regeneration. Among various kind of cytokinins, kinetin was the most effective plant growth regulator for shoot regeneration from root tissues of sweet potatoes. Two types of shoot formation were occurred; the direct shoot regeneration from excised roots without callus formation and the shoot regeneration with a tissue of hard callus-like texture from intact roots grown in vitro. Kinetin of 2.5 - 5.0 ¥ìM and 150 ¥ìM concentrations were required for the best shoot regeneration from excised root and intact root explants, respectively. Shoot formation via hard callus-like tissue may provide possibility that optimization of culture conditions leads to formation of storage roots in vitro.
